What did the DHARMA Institution in Lost originally want from the island?

Like, the DHARMA Institution in "Lost" had to go to the island for a reason, and even if they crashed on the island, what were they doing all that time between them crashing and the plane wreck survivors arriving. And do the numbers: 4,8,15,16,23,and 42 mean anything to you? Thanks!
